Nectin-4(PVRL4)isatypeItransmembraneglycoproteinwhichbelongstothenectinfamilyofIgsuperfamilyproteins.ItcontainstwoIg-likeC2-typedomainsandoneIg-likeV-typedomain.PVRL4seemstobeinvolvedincelladhesionthroughtrans-homophilicand-heterophilicinteractions,thelatterincludingspecificallyinteractionswithnectin-1.Itdoesnotactasreceptorforalpha-herpesvirusentryintocells.Itispredominantlyexpressedinplacenta,theembryoandbreastcarcinoma.Butitisnotdetectedinnormalbreastepithelium.Thesolubleformisproducedbyproteolyticcleavageatthecellsurface(shedding),probablybyADAM17.Mutationsinthisgenearethecauseofectodermaldysplasia-syndactylysyndrometype 1, an autosomal recessive disorder.
