Prolactin (PRL) is a secreted neuroendocrine pituitary hormone that acts primarily on the mammary gland to promote lactation, but has pleiotropic effects in both males and females. Non-glycosylated prolactin is produced by the pituitary and packaged in storage granules before secretion, while glycosylated prolactin is reported to be constitutively secreted, have lower biological potency, and be removed from the circulation more quickly. Prolactin is synthesized mainly by the anterior pituitary in all mammals, where secretion is under tonic inhibition by hypothalamic dopamine. In humans, prolactin is also produced peripherally. Prolactin expression is low during early human pregnancy, but increases in late pregnancy. The prolactin receptor (PRLR) is a transmembrane type I glycoprotein that belongs to the cytokine hematopoietic receptor family. prolactin molecule is thought to bind two receptor molecules. In addition to its lactogenic activity, peripherally produced prolactin plays roles in breast and prostate cancer development, regulation of reproductive function, and immunoregulation.
Phosphinothricin N-acetyltransferase (PAT) is an enzyme that acetylates the free NH2 group of L-phosphinothricin (L-PPT) in the presence of acetyl-CoA as a co-substrate. It is highly specific for L-PPT and does not acetylate other L-amino acids or structurally similar molecules. L-PPT is a glutamate analog that can inhibit glutamine synthetase activity in plants, resulting in the accumulation of ammonia to toxic levels and impairment of photosynthesis. The introduction of a PAT gene into a plant genome can confer resistance to glufosinate herbicide during post-emergent applications.
Growth factor of the TGF-beta superfamily that plays essential roles in many developmental processes, including neurogenesis, vascular development, angiogenesis and osteogenesis. Acts in concert with PTHLH/PTHRP to stimulate ductal outgrowth during embryonic mammary development and to inhibit hair follicle induction. Initiates the canonical BMP signaling cascade by associating with type I receptor BMPR1A and type II receptor BMPR2. Once all three components are bound together in a complex at the cell surface, BMPR2 phosphorylates and activates BMPR1A. In turn, BMPR1A propagates signal by phosphorylating SMAD1/5/8 that travel to the nucleus and act as activators and repressors of transcription of target genes. Can also signal through non-canonical BMP pathways such as ERK/MAP kinase, PI3K/Akt or SRC cascades. For example, induces SRC phosphorylation which, in turn, activates VEGFR2, leading to an angiogenic response.
The chemokine (C-C motif) ligand 2 (CCL2), also known as monocyte chemoattractant protein (MCP)-1 and small inducible cytokine A2 (SCYA2)), is a small cytokine that belongs to the CC chemokine family responsible for monocyte attraction. Its cognate receptor, CCR2, play a critical role in regulating nociceptive processes during neuropathic pain. Both CCL2 and CCR2 are implicated in induction of autoimmunity. CCL2 recruits monocytes, memory T cells, and dendritic cells to the sites of inflammation produced by either tissue injury or infection. Recently research also showed that CCL2 might be useful as a biomarker of fibrosis as well as a target for therapeutic intervention.
Human C-X-C Motif Chemokine Ligand 10 (CXCL10) is a non-ELR chemokine secreted by various cell types; such as monocytes; endothelial cells and fibroblasts; in response to IFN-γ. CXCL10 functions via chemokine receptor CXCR3. CXCL10 has been attributed to several roles; such as chemoattraction for activated T-lymphocytes; inhibition of angiogenesis; and antitumor activity.
