Ephrin type-B receptor 2(EPHB2) belongs to the protein kinase superfamily and Ephrin receptor subfamily. EPHB2 contains 1 Eph LBD domain; 2 fibronectin type-III domains; 1 protein kinase domain and 1 SAM domain. Ephrin receptors and their ligands; the ephrins; mediate numerous developmental processes; particularly in the nervous system. Based on their structures and sequence relationships; ephrins are divided into the ephrin-A (EFNA) class; which are anchored to the membrane by a glycosylphosphatidylinositol linkage; and the ephrin-B (EFNB) class; which are transmembrane proteins. The Eph family of receptors are divided into 2 groups based on the similarity of their extracellular domain sequences and their affinities for binding ephrin-A and ephrin-B ligands. Ephrin receptors make up the largest subgroup of the receptor tyrosine kinase (RTK) family.
Ephrin type-B receptor 2; also known as EphB2; belongs to the ephrin receptor subfamily of the protein-tyrosine kinase family which 16 known receptors (14 found in mammals) are involved: EPHA1; EPHA2; EPHA3; EPHA4; EPHA5; EPHA6; EPHA7; EPHA8; EPHA9; EPHA10; EPHB1; EPHB2; EPHB3; EPHB4; EPHB5; EPHB6. EphB2 receptor tyrosine kinase phosphorylates syndecan-2 and that this phosphorylation event is crucial for syndecan-2 clustering and spine formation. The Eph family of receptor tyrosine kinases (comprising EphA and EphB receptors) has been implicated in synapse formation and the regulation of synaptic function and plasticity6. Ephrin receptors are components of cell signalling pathways involved in animal growth and development; forming the largest sub-family of receptor tyrosine kinases (RTKs). Ligand-mediated activation of Ephs induce various important downstream effects and Eph receptors have been studied for their potential roles in the development of cancer. EphB receptor tyrosine kinases are enriched at synapses; suggesting that these receptors play a role in synapse formation or function. We find that EphrinB binding to EphB induces a direct interaction of EphB with NMDA-type glutamate receptors. This interaction occurs at the cell surface and is mediated by the extracellular regions of the two receptors; but does not require the kinase activity of EphB.

Furthermore; EphB4 protein and the corresponding ligand Ephrin-B2 contribute to tumor growth in various human tumors. EphB4 protein has tumor suppressor activities and that regulation of cell proliferation; extracellular matrix remodeling; and invasive potential are important mechanisms of tumor suppression. Therefore; Ephrin-B2/EphB4 may be recognized as a novel prognostic indicator for cancers.

Ephrin receptors make up the largest subgroup of the receptor tyrosine kinase (RTK) family. EphB6 is an unusual Eph receptor; lacking catalytic capacity due to alterations in its kinase domain. Interestingly; increased metastatic activity is associated with reduced EphB6 receptor expression in several tumor types; including breast cancer. This emphasizes the potential of EphB6 to act as a suppressor of cancer aggressiveness. EphB6 suppress cancer invasiveness through c-Cbl-dependent signaling; morphologic changes; and cell attachment and indicate that EphB6 may represent a useful prognostic marker and a promising target for therapeutic approaches. EphB6 can both positively and negatively regulate cell adhesion and migration; and suggest that tyrosine phosphorylation of the receptor by an Src family kinase acts as the molecular switch for the functional transition. In addition; Ephrin-B2 may be a physiological ligand for the EphB6 receptor.
Ephrin-A1 is a member of the A-type ephrin family of cell surface proteins that function as ligands for the A-type Eph receptor tyrosine kinase family. Ephrin-A1 can be induced by TNF and IL1B. Its expression levels can be down-regulated in primary glioma tissues compared to the normal tissues. The soluble monomeric form is expressed in the glioblastoma multiforme (GBM) and breast cancer cells. Soluble Ephrin-A1 is necessary for the transformation of HeLa and SK-BR3 cells and participates in the relocalization of EPHA2 away from sites of cell-cell contact during transformation. Ephrin-A1 plays an important role in angiogenesis and tumor neovascularization.
