Descrizione Azione

LRG1 (Leucine-Rich Alpha-2-Glycoprotein 1) is a Protein Coding gene. LRG1 belongs to the leucine-rich repeat (LRR) family. Members of this family are involved in protein-protein interaction, signal transduction, and cell adhesion and development. LRG1 is expressed during granulocyte differentiation. It contains 4 LIM zinc-binding domains and 1 Rho-GAP domain. LRG1 is involved in promoting neovascularization (new blood vessel growth) by causing a switch in transforming growth factor-beta (TGFbeta) signaling in endothelial cells. LRG1 binds to the accessory receptor endoglin and promotes signaling via the ALK1-Smad1/5/8 pathway. It may be a potential therapeutic target for the treatment of diseases where there is aberrant neovascularization. Diseases associated with LRG1 include Appendicitis and Normal Pressure Hydrocephalus.

Various members of the low-density lipoprotein receptor (LDLR) family have been reported to play a role in APP trafficking and processing and are important risk factors in AD.LDLR-related protein 1 (LRP1) shuttles between the trans-Golgi Network (TGN); plasma membrane (PM); and endosomes.LRP1 is a functional APP receptor involved in APP trafficking and processing. LRP1 interacts directly with the ectodomain of APP and colocalizes with APP at the TGN. LRP1 is a novel APP sorting receptor that protects APP from amyloidogenic processing; suggesting that a decrease in LRP1 function may contribute to the pathogenesis of Alzheimer's disease.

Receptor-associated protein (RAP) is a molecular chaperone for low density lipoprotein receptor-related protein (LRP), which plays a key role in cholesterol metabolism. The lipoprotein receptor-related protein (LRP) is an endocytic receptor for several ligands, such as alpha2-macroglobulin (alpha2 M) and apolipoprotein E. LRP is involved in the clearance of lipids from the bloodstream and is expressed in the atherosclerotic plaque. The LRP-associated protein (LRPAP in humans, RAP in mice) acts as a chaperone protein, stabilizing the nascent LRP peptide in the endoplasmic reticulum and Golgi complex. Alpha-2-macroglobulin receptor-associated protein, also known as low density lipoprotein receptor-related protein-associated protein 1, RAP and LRPAP1, is a 39 kDa protein and a member of the alpha-2-MRAP family. It is a receptor antagonist that interacts with several members of the low density lipoprotein (LDL) receptor gene family. Upon binding to these receptors, LRPAP1 inhibits all ligand interactions with the receptors. LRPAP1 is present on cell surface forming a complex with the alpha-2-macroglobulin receptor heavy and light chains. It binds with LRP1B and the binding is followed by internalization and degradation. LRPAP1 interacts with LRP1/alpha-2-macroglobulin receptor and LRP2, and may be involved in the pathogenesis of membrane glomerular nephritis. LRPAP1 together with LRP2 forms the Heymann nephritis antigenic complex. LRP2 is expressed in epithelial cells of the thyroid, where it can serve as a receptor for the protein thyroglobulin.

LRRC32 (Leucine-Rich Repeat Containing 32) is a Protein Coding gene. This gene encodes a type I membrane protein which contains 20 leucine-rich repeats. LRRC32, also known as Glycoprotein A Repetitions Predominant (GARP), has been postulated as a novel surface marker of activated T(regs). LRRC32 is a T(reg)-specific receptor that binds latent TGF-beta and dominantly controls FOXP3 and the regulatory phenotype via a positive feedback loop. It belongs to the LRRC32/LRRC33 family and is broadly expressed in the placenta, lung, and other tissues. Alterations in the chromosomal region 11q13-11q14 are involved in several pathologies. An important paralog of this gene is NRROS.
