B7-H3 is a member of the B7 family of immune regulatory ligands that is thought to attenuate peripheral immune responses through co-inhibition. It plays an important role in adaptive immune responses, and was shown to either promote or inhibit T-cell responses in various experimental systems. B7-H3 may play an important role in muscle-immune interactions, providing further evidence of the active role of muscle cells in local immunoregulatory processes. B7-H3 is a novel protein structurally related to the B7 family of ligands by the presence of a single set of immunoglobulin-V-like and immunoglobulin-C-like (VC) domains. Previous studies have correlated its overexpression with poor prognosis and decreased tumor-infiltrating lymphocytes in various carcinomas including uterine endometrioid carcinomas, and mounting evidence supports an immuno-inhibitory role in ovarian cancer prognosis. Recently, B7-H3 expression has been reported in several human cancers indicating an additional function of B7-H3 as a regulator of antitumor immunity.
CD276, also known as B7-H3, is a member of the B7 superfamily with signature IgV and IgG regions in extracellular domains. It is a type I transmembrane protein and shares 20–27% amino acid identity with other B7 family members. B7-H3 is involved in the activation of T lymphocytes, and regulates murine bone formation. It is also reported that B7-H3 may play an important role in muscle-immune interactions, providing further evidence of the active role of muscle cells in local immunoregulatory processes. B7-H3 is expressed on T-cells, natural killer cells, and antigen presenting cells, as well as some non-immune cells, such as osteoblasts, fibroblasts, fibroblast-like synoviocytes and epithelial cells. High expression of B7-H3 in tumor vasculature also correlates with poor survival in patients, suggesting that it may play a role in tumor cell migration.
V-set domain-containing T-cell activation inhibitor 1, also known as B7X, B7H4, B7S1, and VTCN1, is a single-pass type-III membrane protein belonging to the B7 family of costimulatory proteins. These proteins are expressed on the surface of antigen-presenting cells and interact with ligands on T lymphocytes. They provide costimulatory signals that regulate T cell responses. A soluble form of B7H4 has also been detected. B7X / VTCN1 / B7H4 negatively regulates T-cell-mediated immune response by inhibiting T-cell activation, proliferation, cytokine production and development of cytotoxicity. When expressed on the cell surface of tumor macrophages, B7X / VTCN1 / B7H4 plays an important role, together with regulatory T-cells(Treg), in the suppression of tumor-associated antigen-specific T-cell immunity. B7X / VTCN1 / B7H4 is also involved in promoting epithelial cell transformation. This membrane protein can be up-regulated by IL6 / interleukin-6 and IL10 / interleukin-10 and inhibited by CSF2 / GM-CSF and IL4 / interleukin-4 on antigen-presenting cells.
B7 Homolog 4 (B7-H4) is glycosylated member of the B7 family of immune costimulatory proteins. Mature human B7-H4 consists of a 235 amino acid (aa) extracellular domain (ECD) with two Ig-like V-type domains; a 21 aa transmembrane segment; and a 2 aa cytoplasmic tail. It is widely expressed; including in kidney; liver; lung; pancreas; placenta; prostate; spleen; testis and thymus. B7-H4 negatively regulates T-cell-mediated immune response by inhibiting T-cell activation; proliferation; cytokine production and development of cytotoxicity. When expressed on the cell surface of tumor macrophages; plays an important role; together with regulatory T-cells (Treg); in the suppression of tumor-associated antigen-specific T-cell immunity. It also involved in promoting epithelial cell transformation.
