Pectinase is a class of pectin decomposition enzymes, including propectinase, pectin esterase, polygalacturonase and pectin lyase, which widely exist in plant fruits and microorganisms, and is the most important enzyme in fruit processing. The detection principle of this kit is that the substance generated by the hydrolysis of pectin by pectinase reacts with the chromogenic agent to produce a brown-red substance with characteristic absorption peak at 540 nm. The activity of pectinase can be calculated by measuring the change of absorbance value at 540 nm.

Pepsin can catalyze substrate hydrolysis, and the hydrolysate reacts with the chromogenic agent under alkaline conditions to form a blue substance. The blue substance has a maximum absorption peak at 660 nm, and the color depth is proportional to the activity of pepsin. The enzyme activity can be calculated by OD value

Plant peroxidase, a member of the superfamily of peroxidase, catalyzes the redox reaction between H2O2 and various reductants. The plant peroxidase has the same general structure and consists of iron porphyrin IX and ten α-helixes. Based on the difference of primary structure, the superfamily of plant peroxidase can be divided into three types: class I (intracellular type), class II (extracellular type of fungi) and class III (secreted type of plant).
