The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human GHR. You can calculate the concentration of Human GHR in the samples by comparing the OD of the samples to the standard curve.

During the reaction, Human GHRL in the sample or standard competes with a fixed amount of Human GHRL on the solid phase supporter for sites on the Biotinylated Detection Ab specific to Human GHRL. Excess conjugate and unbound sample or standard are washed away, and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns from blue to yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of Human GHRL in tested samples can be calculated by comparing the OD of the samples to the standard curve.

Samples or Standards are added to the micro CLIA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human GIF and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human GIF, biotinylated detection antibody and Avidin-HRP conjugate will appear luminescence. The Relative light unit (RLU) value is measured by the Chemiluminescence immunoassay analyzer at 425nm. The RLU value is positively associated with the concentration of Human GIF. You can calculate the concentration of Human GIF in the samples by comparing the RLU value of the samples to the standard curve.
