The cluster of differentiation (CD) system is commonly used as cell markers in immunophynotyping. Different kinds of cells in the immune system can be identified through the surface CD molecules which associating with the immune function of the cell. There are more than 320 CD unique clusters and subclusters have been identified. Some of the CD molecules serve as receptors or ligands important to the cell through initiating a signal cascade which then alter the behavior of the cell. Some CD proteins do not take part in cell signal process but have other functions such as cell adhesion. CD180, also known as RP105, is a B-cell surface molecule belonging to the family of pathogen receptors, Toll-like receptors (TLR). CD180 has an extracellular leucine-rich repeats and a short cytoplasmic tail. CD180 / RP105 interact with an extracellular molecule named MD1 and then together form the cell surface receptor complex RP105 / MD1 which induces B-cell activation in humans and mice, leading to proliferation and up-regulation of a costimulatory molecule, B7.2 / CD86. CD180 / RP105 also has a role in LPS response because B cells lacking RP105 show hyporesponsiveness to LPS.
GAMT is a methyltransferase which belongs to the class I-like SAM-binding methyltransferase superfamily. It contains one RMT2 (arginine N-methyltransferase 2-like) domain and is expressed in liver. GAMT converts guanidoacetate to creatine, using S-adenosylmethionine as the methyl donor. Defects in GAMT are the cause of guanidinoacetate methyltransferase deficiency, which is an autosomal recessive disorder characterized by developmental delay/regression, mental retardation, severe disturbance of expressive and cognitive speech, intractable seizures and movement disturbances, severe depletion of creatine/phosphocreatine in the brain, and accumulation of guanidinoacetic acid in brain and body fluids.
Gankyrin is a multicatalytic proteinase oncoprotein consists of 7 ankyrin repeats. Gankyrin overexpressed in most hepatocellular carcinomas. Gankyrin is involved in theregulation of the phosphorylation of the retinoblastoma protein by CDK4 to enhance the ubiquitinylation of p53 by MDM2. Gankyrin is also involved in progression of esophageal squamous cell carcinoma. Gankyrin plays an oncogenic role especially in early stages of human epatocarcinogenesis.
Neuromodulin, also known as Axonal membrane protein GAP-43, Growth-associated protein 43, Neural phosphoprotein B-50, pp46 and GAP43, is a cell membrane protein which belongs to theneuromodulin family. Neuromodulin / GAP43 contains oneIQ domain. Neuromodulin / GAP43 is associated with nerve growth. It is a major component of the motile "growth cones" that form the tips of elongating axons. Neuromodulin / GAP43 is involved in neurite outgrowth, a crucial process for the differentiation of neurons. The sudden infant death syndrome (SIDS) is the main cause of postneonatal infant death and its cause is still unknown. Neuromodulin / GAP43 is a marker of synaptic plasticity and is critical for normal development of the serotonergic innervation. Neuromodulin / GAP43 is a major cortical cytoskeleton-associated and calmodulin binding protein that is widely and abundantly expressed during development, maintained in selected brain structures in the adult, and reinduced during nerve regeneration. CAP23 and GAP43 are functionally related intrinsic determinants of anatomical plasticity. These proteins function by locally promoting subplasmalemmal actin cytoskeleton accumulation.
