DMRT1, also named as DMT1, belongs to the DMRT family. DMRT1 is one of the most conserved transcription factors in a variety of species and is involved in sex determination and germ cell differentiation. DMRT1 prevents sexual switch from male to female, represses pluripotency, and balances mitosis versus meiosis. DMRT1 also maintains spermatogenesis and replenishes mGSCs by collaborating with Plzf and Sal-like protein 4.
Enables protein-folding chaperone binding activity and unfolded protein binding activity. Involved in several processes, including negative regulation of endothelial cell migration; negative regulation of inclusion body assembly; and protein refolding. Located in cytosol and membrane.
DNAJC6, also named AUXILIN, belongs to the evolutionarily conserved DNAJ/HSP40 family of proteins, which regulate molecular chaperone activity by stimulating ATPase activity. It is a neuronal protein that functions specifically in the pathway of clathrin-mediated endocytosis. It shares homology with GAK, and the two proteins act as cochaperones to support the HSC70 -dependent clathrin uncoating of clathrin-coated vesicles (PMID: 2016009). DNAJC6 is expressed in various brain regions, including cerebellum, corpus callosum, cortex, striatum, brainstem, pons, putamen, spinal cord and substantia nigra.
This gene encodes the catalytic subunit of the DNA-dependent protein kinase (DNA-PK). It functions with the Ku70/Ku80 heterodimer protein in DNA double strand break repair and recombination. The protein encoded is a member of the PI3/PI4-kinase family.
This gene encodes an enzyme that transfers methyl groups to cytosine nucleotides of genomic DNA. This protein is the major enzyme responsible for maintaining methylation patterns following DNA replication and shows a preference for hemi-methylated DNA. Methylation of DNA is an important component of mammalian epigenetic gene regulation. Aberrant methylation patterns are found in human tumors and associated with developmental abnormalities. Variation in this gene has been associated with cerebellar ataxia, deafness, and narcolepsy, and neuropathy, hereditary sensory, type IE. Alternative splicing results in multiple transcript variants.

CpG methylation is an epigenetic modification that is important for embryonic development, imprinting, and X-chromosome inactivation. Studies in mice have demonstrated that DNA methylation is required for mammalian development. This gene encodes a DNA methyltransferase that is thought to function in de novo methylation, rather than maintenance methylation. The protein localizes to the cytoplasm and nucleus and its expression is developmentally regulated.
